Yes, but beware of the limitation of the ISO-9660
format: at most 64k "parent" directories could exist
on a ISO-9660 CD or DVD. In fact, depending on the
structure of your tree they could be well under 64k
"parent" directories. And with 4+ GB you will quick
stumble across it. 

mkisofs will gently remind you and refuse to prepare
the image ...
